 DNAは生命現象に関わる基本的な物質として、盛んに研究が行われてきています。DNAの塩基配列や原子配列については、近年、飛躍的な研究の進展があり、現代の生命科学の中心課題ともなっています。一方で、細胞由来のDNA分子は、その全長は、mmからcm程度の大きさがあり、それが全体としてどのような特質を持っているかといったことについては、不明な点が多く残されています。我々の研究室では、蛍光顕微鏡を活用して、長鎖DNAの単分子観察を行うことにより、溶液環境の変化にともなう、DNAの高次構造変化について系統的な研究を進めてきました。このような研究を通した、巨大DNA1分子は大きく広がったランダムコイル状態から、高密度に凝縮した状態へと、不連続な転移を示すことを明らかにしてきました。この転移では密度差が、数万倍程度であり、構造的にも不連続―連続転移で特徴付けられることから、Landau流の対称性の議論に基づくと、一次相転移に分類されます。また、このような単一分子鎖レベルでの、相転移は、DNAのsemiflexible(全体の長さのスケールから見ると柔らかいが、局所的に見ると堅いということ)な性質に起因しており、DNAに限らず高分子の一般的な特質とも関連して、今後の展開が期待される研究分野となっています。  DNA凝縮転移に解明には、学際的なアプローチが求められています。本論文ではDNAの単一分子鎖の実験と、semiflexibleな高分子についての分子動力学による理論計算を、組み合わせて総合的に研究を行った結果を報告しています。  単一DNA分子の実験では、水溶性の中性高分子であるPEGを添加することによる、単分子鎖の折りたたみ転移の観察を行いました。コイル状態から凝縮状態への過程と、その逆過程を計測した結果、明瞭で、再現性のあるヒステリシスがあることを、見出しました。これは、DNA単一分子鎖レベルでのヒステリシスを明確にしてものとして、実質的に最初の論文となっています。  上述の実験結果に、分子鎖のsemiflexibilityがどのように関与しているかといった問題に迫るために、無荷電の分子鎖について、折り曲がりの自由度に硬さ(バネ定数)を導入することにより、分子動力学計算を行いました。分子鎖が柔らかい時にはヒステリシスは生じることは無く、十分な堅さを取り入れたときにのみ、ヒステリシスが現れることを確認しまました。分子鎖の堅さが増すと、折りたたまれた状態の秩序性は高く、このことが、折りたたみ転移が、凝縮と脱凝縮でその経路が異なることに直接関与していることが明らかとなりました。  このように、単一分子鎖の計測実験と、分子動力学計算による理論的なアプローチを併行させることにより、単一長鎖DNAのもつ特質を抽出することができました。 DNA is one of the most studied molecules among the large number of different biomolecules because of its fundamental role in life. Among its molecular properties, the conformational transition of a giant DNA has recently attracted a great deal of attention, because all of the genomic DNA exhibit the size longer than hundreds kilo base-pairs. The research laboratory of Prof. Yoshikawa has pioneered the basic research to elucidate the conformational transition of single DNA molecules caused by condensing agents through single molecular imaging method. As the condensing agents can be multi- valent cations or neutral polymers, such as PEG (polyethylene glycol), are known. This laboratory has revealed the most striking feature of a giant DNA, that is, the long chain DNA undergoes a discrete conformational transition, accompanied with finite region for the coexistence between compact and elongated conformations. The discrete transition is an indication of the first-order phase transition under the criterion of symmetry argument by Landau, and an extensive study has shown that the semiflexibility of DNA is the main cause for such unique behavior. To target on this problem of the phase transition of a single molecular chain, an integrative approach by combining experimental and computational methods was developed in this paper. In the experimental part, the forward and backward titration were conducted to monitor the folding and unfolding processes of a giant DNA by increasing and decreasing PEG concentration, respectively. We find that these two processes form a hysteresis loop in a reproducible manner, suggesting that the folding and unfolding processes of a giant DNA undertake different kinetic pathways. This paper presents the first experiment evidence to characterize the conformational hysteresis loop of a single DNA. In addition, computer simulations were resorted to examine the role of semiflexibility on hysteresis loop by using a simplified chain model. The simulations predict that the hysteresis loop emerges only when the chain is stiff enough, but not in the case of flexible chains. Meanwhile, the compact conformation in the hysteresis loop is found to be more stable in terms of equilibrium thermodynamics. Nevertheless, the elongated DNA chain, which is thermodynamically less stable, persists until the concentration of condensing agents becomes high enough. We discuss such behavior in relation to the semiflexibility of a DNA molecule. Namely, an increase of chain stiffness attenuates the number of chain conformations, including most of conformations required to make a transition to compact conformation. As a result, the folding process is impeded. On the contrary, condensing agents increase monomer-monomer attraction, which facilitate the folding process by increasing the number of chain conformations. Through the integrative approach between single DNA observation and numerical simulation of a semiflexible polymer chain, we mde clear the underlying physics on the conformational hysteresis of DNA. Future study along this direction may open a new discipline on the polymer science not only for DNA but also for macromolecules in general.
